کارِ سعادت
قیاس ذاتی کسی بھی اعتراض گزار کی بنیادی قوت ہوتی ہے جسے وہ شعور (خود ساختہ شعور جو آرٹ کے درجے میں ہے) کے ذریعے کسی معروضی حقیقتِ مطلق کو اپنے شعور کے مطابق قیاس کر کے مسرور ہوتا ہے تبھی کہا جاتا ہے خود سے یگانگی حقیقت سے بیگانگی کے مترادف ہوتی ہے۔ ہرہیچ مدان و ناتواں کو اپنے عقلِ کل ہونے کا زعم وقوف سے عاری کرتا ہے اس لیے مضحکہ خیز اعتراضات کو فتوحات گردانتا ہے۔ حالاں کہ راست علم ہمیشہ کسی واقعی معروض کی تمیز سے اٹھتا ہے یعنی موجود پر قوتِ امتیاز یا وجدانی ادراک کے ذریعے جوہر کے اسما و شناخت کا سفر طے کیا جاتا ہے۔ ہر چند ہر مذہب موضوعِ علم بننے کی پوری استطاعت رکھتا ہے۔ ہاں مگر ناظر کاصاحبِ وجدان ہونا ازحد ضروری ہوتا ہے۔ چوں کہ ہر قضیہ وجدانی ادراک کے متحرک ہونے سے جنم لیتا ہے۔ ہم زمان و مکان کا ادراک اسی استعداد کی تحریک سے کرتے ہیں۔ اسی لیے ایک طے شدہ امر ہے کہ شعور ِ علمی کی تشکیل میں خارجی معروض کا ہونا بنیادی حیثیت کا حامل ہے۔ اگر خارجی معروض اور اس سے منسوب شعورِ انسانی میں مطابقت نہ ہو تو قضیہ کے بجائے بے بنیاد مناقشات ظہور کرتے ہیں ،جن پہ خارجی واقعیت دلالت نہیں کرتی اور نتیجہ خیز حقانیت کی راہیں بھی برابر مسدود ہوتی جاتی ہیں۔ جب معترضین اپنے خود ساختہ علم کا قابلِ قبول شعور پیدا کرنے سے قاصر ہو جاتے ہیں تو اس کی غلط توجیہات پر قانع ہونے کے شعور سے بھی ہاتھ دھو بیٹھتے ہیں۔ اکثر یوں ہوتا ہے کہ نظریاتی تسکین جب بھی ضرورت سے زیادہ بڑھ جاتی ہے تو معیاری اور یقینی علم کو محال کر دیتی ہے۔

Youth is an integral part of any human society, particularly in an Islamic society. Youth is considered as a junction of past, present and future. Youth can play a positive and effective role against various evils which are prevalent in human society. Important aspects of utilization of youth may include the role of youth in religious, social, political, ethical and educational fields and in restructuring of society. All of these aspects have been discussed in this article, in the light of Islam. Moreover, it is an effort to motivate the youth for practical actions. For the well-being and progress of Muslim Youth and human society adoption of following measures are important and necessary: 1. To gain knowledge of the Revealed message (Shariah) and act upon it. 2. To gain knowledge from true and sincere Scholars (Ahl-eHaqq). 3. Self-accountability, which consists of following points: § Islah of Aqeedah and ideology § To perform good deeds (Amal-e-Salih) § Sincere repentance § Consciousness about purpose of life § Feeling gratified on adopting Islamic Shariah. 4. To have love or hatred only for sake of Allah.

Mapping Genes Causing Syndromic and Non- Syndromic Human Hereditary Skin Disorders

Genetic defects in complex processes of embryonic development and postnatal maintenance of the skin and its appendages result in clinically and genetically heterogeneous group of skin disorders. Due to nonspecific presentation, variable clinical manifestations and highly overlapping phenotypes, the diagnosis of skin disorders is a challenging job for the clinicians and geneticists. However, recent advances in molecular biology technologies notably whole exome sequencing (WES) and microarray have incredibly accelerated identification of genes involved in inherited diseases. The research work, presented in this dissertation, described clinical and molecular characterization of seventeen families of Pakistani origin (A-Q) segregating various forms of syndromic and non-syndromic skin disorders. This included one with palmoplantar hyperkeratosis (A), three with nail disorders (B-D), one with xeroderma pigmentosum (E), two with non-photosensitive trichothiodystrophy and mitral regurgitation (F-G), three with different types of ichthyosis (H-J), two with epidermolysis bullosa (K-L), and five with different types of hair abnormalities (M-Q). In all seventeen families, combination of at least two or three techniques, including microsatellite/SNP genotyping and Sanger/Exome sequencing, led to the establishment of linkage on human chromosomes and detection of potential disease causing variants in different genes. This included fourteen novel variants in 13 different genes (SLURP1, SLCO2A1, ERRC5, MPLKIP, KRT83, ALDH3A2, ABHD5, PLEC, BTAF1, C3orf52, MTUS1, SGSM1, DCAF1) and previously reported three variants in 3 genes (RSPO4, FZD6, KRT14). Disease causing variants, identified in six genes (KRT83, SLCO2A1, BTAF1, MTUS1, C3orf52, SGSM1), are the first report of involvement of such genes in causing skin disorders. Pathogenicity of disease causing variants were tested and verified by various bioinformatics tools (SIFT, PolyPhen, MutationTaster, MutationAssessor, GERP++, phyloP). Non-polymorphic nature of the variants was validated by screening large number of ethnically matched control individuals and by searching various databases. In a couple of cases, qPCR was used to monitor effect of the variants on expression of other genes.
